چکیده

Glutamatergic system stimulationthe nucleus accumbens shell, may affect anxiety-related behaviors, aversive learning and memory. Glutamate receptors are differentially distributed in preand postsynaptic sites contributing to neuronal communications. The present study aimed to examine the possible involvement of the NAc shell presynaptic NMDA receptors on NMDA induced responses, using the elevated-plus maze (EPM) task in maleWistar rats. Bilateral guide cannulae were implanted to allow microinjection of glutamatergic agonist (NMDA) or ca +2 channel blocker (SKF96365 hydrochloride) agents. Pretest intra-NAc shell infusion of NMDA induced anxiolytic-like behaviors and impaired the EPM-associated memory upon test and retest, respectively. In addition our findings showed that, the intra-NAc shell infusion of Ca +2 channel blocker at applied doses, does not alter the anxiety-like response and aversive memory upon test and retest, respectively. Furthermore, infusing the subthreshold dose SKF prior to the injection of effective doses of NMDA, reduced the anxiolytic-like response and improved the aversive memory impairment which had already been induced by intra-NAc shell NMDA injection. Our study showed that, inhibition of the neurotransmitter exocytosis from pre-synaptic neuron via Ca +2 channel blockade bySKF96365 decreases affected induced by NMDA in the NAc shell region, indicating the involvement of the pre-synaptic NMDA receptors in NMDA induced responses. Therefore, NMDA's ability to increase anxiolytic-like behaviors and the aversive memory impairment may be the result of an action on pre-synaptic glutamatergic receptors which in turn decrease the glutamate effect at synaptic terminal level.

Anxiolytic-like Effects and Increase in Locomotor Activity Induced by Infusions of NMDA into the Ventral Hippocampus in Rat: Interaction with GABAergic System

Introduction: In this study, we investigated the role of N-Methyl-D-Aspartate (NMDA) receptors in the ventral hippocampus (VH) and their possible interactions with GABAA system on anxiety-like behaviors. Methods: We used an elevated-plus maze test (EPM) to assess anxiety-like behaviors and locomotor activity in male Wistar rats. Results: The results showed that intra-VH infusions of diffe...

Glutamate Receptors in Nucleus Accumbens Can Modulate Canabinoid-Induced Antinociception in Rat’s Basolateral Amygdala

Introduction: It has been shown that administration of WIN55,212-2, a cannabinoid receptor agonist, into the basolateral amygdala (BLA), dose-dependently increases the thermal latency to withdrawal in the tail-.ick test and decreases pain related behaviors in both phases of the formalin test. Recent human and animal imaging data suggest that the nucleus accumbens (NAc) is an important neural su...

Involvement of D₁/D₂ dopamine antagonists upon open-arms exploratory behaviours induced by intra-nucleus accumbens shell administration of N-methyl-D-aspartate.

Glutamatergic system stimulation in some parts of the brain may affect anxiety-related behaviours, aversive learning and memory. This system retains many interactions with dopaminergic neurotransmission. We have studied the effect of nucleus accumbens (NAc) shell glutamatergic system activation on anxiety-related behaviours as well as aversive learning and memory in adult male Wistar rats using...

The antinociceptive effect of 17β-estradiol in the nucleus paragigantocellularis lateralis of male rats may be mediated by the NMDA receptors

Introduction: The nucleus paragigantocellularis lateralis (LPGi) is involved in the descending pain modulation. The neurostreoid, 17β-estradiol found in the PGi nucleus and modulates nociception by binding to estrogen receptors and also by allosteric interaction with NMDA receptors. In this study, the role of NMDA receptors in the 17β-estradiol-induced pain modulation was investig...

Ameliorative activity of co-administration of ketamine and ECT on depression like behaviors in depressed rats

Background and Objective: Electroconvulsive therapy (ECT) is the end choice treatment for patients with major depressive disorder (MDD). Regarding researches that show NMDA receptor inactivation could yield the same results as anti-depressant drugs, however, the present study examined co-administration of ECT and ketamine (as NMDA antagonist) on depressed rat behaviors.   Materials and Methods...
